These are the profiles of Seta Soujiro and Makimachi Misao. There are spoilers in here, so you are warned.
Soujiro
Name: Seta Soujiro
Nickname: Tenken no Soujiro, kiddo.
Age: 18
Birthday: September 1861
Technique: Tenbu No Sai Niyoru Ken. (Tenken for short.)
Blood Type: AB
Height: 5ft 4 inches.
Weight: 51kg (about 112 pounds.)
Soujiro’s personality is that of a false, cheerful appearance. An ever-constant smile hangs on his lips, and has been that way since he joined Makoto Shishio at the age of eight. He is polite almost to a fault, and a cool, calm exterior constantly surrounds him. Unfortunately, he seems to be devoid of all emotions except unvarying happiness. But be wary...don't judge a book by it's cover.
History:
This is perhaps, along with Himura Kenshin’s story, the most ill fated past in Rurouni Kenshin. Seta Soujiro was the son of a prostitute, and was given to her only relatives for his own good. His adoptive 'parents' of course wanted nothing to do with the child; he was beaten on a frequent basis for their entertainment. Raised as something lower than a slave, he was forced to do hard labor from sun up to sun down. One night, he hears a sound and goes to investigate.
What greets him is the burned man named Makoto Shishio. Soujiro was spared being killed by the smile on his lips, and Shishio lets him live in return of bandages and food. While there, Shishio first presents Soujiro with his words, “The strong live, the weak die” along with a small Wakizashi. Soon his family finds out, and goes out to murder Soujiro. Soujiro in turn flees for his life, finding the Wakizashi and killing his adoptive family out of fright. In the rain, his lips are curled into a smile, while the rain droplets camouflage the tears on his face. From that night on, he was trained to be the right hand man of Makoto Shishio and his group, The Juppongattana.
Journey to a new life.
To make a long story short, Kenshin ends up defeating Soujiro. After his defeat, Soujiro is now capable of emotion which he had locked up for years. Like Kenshin, Soujiro vows to never draw blood again, and sets off. The last we see of him (for he made no more appearances throughout the manga and anime) he is heading along a road, a true smile on his face and a re-newed and willing mind. What ends up of him is up to the fans, and clearly, a lot of them tend to lean toward him eventually meeting Misao. Some go along the lines of falling in love, or creating a sort of triangle between Soujiro, Misao, and Aoshi.

Name: Makimachi Misao.
Nickname: Weasel Girl.
Age: 16
Birthday: November 1863
Technique: Kempo
Blood Type: B
Height: 4ft 10 inches.
Weight: 37kg (81 pounds.)
Personality:
She is perhaps, the happiest and bounciest character in Rurouni Kenshin. Always positive and friendly, it’s hard not to like her. She befriends Kenshin while in Kyoto, as well as Kaoru and the rest of the group. She never seems to loose energy, and is quite active for a sixteen year old. Although she isn’t all fun and smiles, she, like all the other characters, has her depressing points and a deeper look into her persona.
History:
The origin of her parents are unknown; but what is known about her is that she, at the age of six or so was left by Aoshi with her adoptive grandfather Okina. She was raised and trained as an Oniwabanshu ninja, and is quite skilled. She seems to have a crush on Aoshi, although it’s more like sisterly admiration than anything else. At the age of sixteen, she set off on her own throughout Japan in search of the other Oniwabanshu, Hanya, Beshimi, Hyotoko, Shikijo and Aoshi. Along the way she meets up with Kenshin, and is returned. Sadly Kaoru is the bearer of bad news, and informs the awe-struck Misao that everyone excluding Aoshi are dead. A lot of things happen during the Kyoto arc involving her, such as her fight with Kamatari.
She also appears in both the Jinchuu arc, and the final ending of RK, Kenshin Kaden. In the Jinchuu she is the same, except that her admiration toward Aoshi has simmered down. By the Kenshin Kaden however, all trace of that admiration has apparently vanished. Despite the various rumors spread about the Kenshin Kaden, Aoshi and Misao do not in fact, get married in the end.
Possibilities?
So, what exactly makes it such a fun idea to pair Misao up with Soujiro? One reason might be because they’re closer to the same age, as well as similar body builds and happy smiles. Looking deeper, one might find the two to be greatly similar, as many of SouMi or RK fans claim to see. Because she has such a vibrant, sociable personality, it is quite easy to see her warming up to someone like Soujiro. He is, after all very much like Kenshin, and they get along together just fine. It is however up to the fans, and plainly seen, a lot of people pair these two together!
